SMP-656 is a HER2-targeting antibody-drug conjugate (ADC) developed using the XL-XDC platform by Xiling Lab. It utilizes eribulin as its cytotoxic payload, which acts as a tubulin inhibitor, and is linked to the antibody via a proprietary SuperHydro linker designed to enhance stability and payload delivery. The drug aims to overcome resistance seen with other approved ADCs or cytotoxic agents targeting HER2. SMP-656 is currently in Phase 1 clinical trials for advanced solid tumors expressing or mutated HER2.
