Overview

SMRX-11, also known as Clot Specific Streptokinase (CSSK), is a recombinant biologic therapeutic protein developed by Symmetrix Biotech (a subsidiary of Nostrum Pharmaceuticals) under license from the Institute of Microbial Technology (IMTECH). It is an engineered plasminogen-activating hybrid protein produced in a *Pichia pastoris* yeast system. The molecule consists of a streptokinase core with fibrin-binding domains attached to both ends, which mask its activity in the systemic circulation. This design allows the drug to remain inactive until it encounters a blood clot, where it binds to fibrin and stimulates the conversion of plasminogen to plasmin. SMRX-11 is intended for the treatment of acute myocardial infarction (AMI) and ST-segment elevation myocardial infarction (STEMI) via rapid bolus injection, offering potential safety advantages such as fibrinogen-sparing properties and reduced bleeding risk compared to conventional thrombolytics.
