Smurf1 inhibitor is a small molecule therapeutic candidate being developed by Interprotein for the treatment of cardiovascular diseases, specifically pulmonary arterial hypertension (PAH). The drug functions by inhibiting SMAD specific E3 ubiquitin protein ligase 1 (Smurf1), an enzyme that mediates the degradation of bone morphogenetic protein (BMP) signaling components, including SMAD1/5 and BMP type II receptors. In PAH, the loss of BMP signaling leads to pathological vascular remodeling; by blocking Smurf1-mediated degradation, this inhibitor aims to restore BMP pathway activity and ameliorate disease progression. The program is currently in the preclinical hit validation stage.
