Overview

SMW139 is a small molecule radiolabeled with carbon-11 ([11C]SMW139) that acts as a high-affinity antagonist (Ki 32 nM) of the P2X7 receptor, a cell surface receptor upregulated in activated microglia. It is primarily developed as a positron emission tomography (PET) tracer to image neuroinflammation, especially for research in neurodegenerative and neuroinflammatory diseases such as multiple sclerosis and potentially Parkinson's disease. SMW139 is characterized by selective P2X7 receptor binding and favorable pharmacokinetics for in vivo imaging. Initial toxicity studies in rats show no adverse effects at microdose levels. Additional clinical validation is ongoing for the assessment of neuroinflammation in humans[1][3][5][7][8].
