Drug intelligence / Profile preview
SN-2310 is an injectable emulsion formulation of SN-38 (7-ethyl-10-hydroxycamptothecin), the active metabolite of the widely used chemotherapeutic irinotecan. Developed by Sonus Pharmaceuticals using its proprietary TOCOSOL vitamin E-based delivery technology, SN-2310 was designed to provide direct systemic delivery of SN-38, thereby bypassing the variable and often inefficient hepatic conversion required for irinotecan. As a potent inhibitor of DNA topoisomerase I, SN-38 stabilizes the cleavable complex between the enzyme and DNA, leading to lethal double-strand DNA breaks during replication. SN-2310 was evaluated in Phase 1 clinical trials for the treatment of advanced solid malignancies, demonstrating an improved safety profile and reduced toxicity compared to irinotecan. However, following the merger of Sonus Pharmaceuticals with OncoGenex (now Achieve Life Sciences), development of the candidate was discontinued.
