SN2310 is a novel, injectable Vitamin E-based emulsion formulation of camptothecin developed by Sonus Pharmaceuticals using its proprietary TOCOSOL drug delivery technology. Camptothecin is a potent cytotoxic alkaloid that inhibits DNA topoisomerase I, an enzyme essential for DNA relaxation during replication and transcription. By stabilizing the covalent topoisomerase I-DNA cleavage complex, camptothecin prevents DNA ligation, leading to double-strand breaks and cell death. The TOCOSOL delivery system was designed to enhance the solubility and stability of the active lactone form of camptothecin, which is otherwise unstable at physiological pH, thereby potentially improving its safety and efficacy profile. SN2310 reached Phase 1 clinical trials for the treatment of advanced solid tumors but development was discontinued following the merger of Sonus with OncoGenex and a strategic shift in the company's pipeline.
