Overview

SN32976 is a small molecule, orally bioavailable inhibitor of the phosphatidylinositol 3-kinase (PI3K) signaling pathway, specifically targeting the PI3K alpha isoform. Developed by Pathway Therapeutics, it exhibits preferential activity against PI3K alpha (IC50 = 28 nM) compared to PI3K delta (IC50 = 287 nM) and mTOR (IC50 = 227 nM). In preclinical studies, SN32976 demonstrated significant anti-tumor activity in human tumor xenograft models, such as the U87 MG glioblastoma model, showing superior efficacy compared to the pan-PI3K inhibitor ZSTK474. Although it was a promising preclinical candidate for cancer treatment, its development was eventually superseded by its more metabolically stable analogue, PWT33597, which progressed into human clinical trials.
