Drug intelligence / Profile preview
SNA043 (also known as SNA043-SmartCC) is a preclinical-stage bispecific fusion protein tetramer developed by SmartNuclide for two-step pretargeted radioimmunotherapy (PRIT) of carbonic anhydrase IX (CA9)-expressing clear cell renal cell carcinoma (ccRCC). Engineered using camelid single-domain antibodies (nanobodies) and antibody engineering, SNA043-SmartCC consists of a tumor-targeting domain directed against CA9, a binding domain for the lanthanide metal chelator SmartD2, and a human-derived tetramerization domain. In the PRIT regimen, SNA043-SmartCC is administered first to target and accumulate in CA9-expressing tumor tissues, followed by the administration of the radiolabeled bivalent chelator 177Lu-SmartD2, which binds to the pretargeted fusion protein to deliver localized radiation while minimizing systemic toxicity.
