Drug intelligence / Profile preview
SNAP-BiTE is a universal bispecific T-cell engager (BiTE) consisting of a fusion of the SNAPtag self-labeling enzyme to an anti-CD3 single-chain variable fragment (scFv). Developed by researchers at the University of Pittsburgh, SNAP-BiTE is designed to be secreted by engineered T cells (including CAR T cells) to retarget T-cell activity. The SNAPtag portion of the fusion protein covalently binds to co-administered antibody 'adaptors' conjugated with a benzylguanine (BG) motif. This modular design allows for post-translational, user-defined targeting of multiple tumor antigens (including HER2, CD20, EGFR, and folate receptor alpha) using clinical-grade antibodies like trastuzumab, rituximab, cetuximab, and mirvetuximab, thereby broadening and amplifying anti-tumor immune responses.
