Overview

SNAP-CAR is a modular chimeric antigen receptor (CAR) technology platform designed to create "universal" CAR T or natural killer (NK) cell therapies for cancer. Unlike conventional CAR-T therapies that are engineered to target a single tumor antigen, the SNAP-CAR system uses a self-labeling enzyme called the SNAP-tag fused to the CAR receptor. This allows therapeutic cells to be redirected against different tumor antigens by co-administering benzylguanine-labeled antibodies specific for those antigens. The antibody binds its target on the tumor and covalently attaches via benzylguanine to the SNAP-tag on the engineered immune cells, forming an active immunotherapeutic complex. This approach enables programmable targeting of multiple or changing cancer antigens and offers tunable potency by adjusting antibody levels. Preclinical studies have shown efficacy in mouse models of both hematologic and solid tumors, with ongoing development focused on HER2-expressing cancers as an initial indication[1][3][4][5][6][8].
