SNF472 is an intravenous formulation of myo-inositol hexaphosphate (phytate) developed as a novel inhibitor of pathological calcification. It acts by selectively binding to hydroxyapatite crystals, thereby inhibiting their formation and growth in vascular tissues without chelating free calcium. This mechanism targets the final common pathway of cardiovascular calcification (CVC) and calcific uremic arteriolopathy (calciphylaxis), particularly in patients with end-stage renal disease undergoing hemodialysis. SNF472 has demonstrated efficacy in preclinical models and clinical trials by reducing progression of coronary artery and aortic valve calcification, improving wound healing, and reducing pain associated with calciphylaxis. The drug is currently in phase 3 development for these indications and has received orphan drug designation from both the FDA and EMA[1][2][3][4][5][6][7][8].
