SNIPER bispecific antibodies are engineered therapeutic antibodies developed by Invenra that simultaneously target two distinct antigens on tumor cells. Each arm of the antibody is designed with low to moderate affinity for its respective antigen (such as GD2 and B7H3), but when both arms engage their targets on the same cell, the overall binding strength (avidity) becomes high. This dual-targeting approach ensures that the antibody binds strongly only to cells expressing both antigens—typically tumor cells—while minimizing binding to healthy tissues that may express only one antigen at low levels. The result is increased specificity for tumor cells and reduced off-target toxicity compared to conventional monoclonal antibodies. Preclinical studies have demonstrated efficacy in models of neuroblastoma and melanoma, with mechanisms including enhanced immune cell engagement (e.g., ADCC) and selective depletion of intratumoral regulatory T cells to modulate the tumor microenvironment[1][3][4][5].
