SNOH-3 is a novel small molecule inhibitor of histone deacetylases (HDACs) with demonstrated activity in both cell-based and animal tumor models. It strongly inhibits HDAC activity, especially HDAC1 and HDAC3, leading to increased histone acetylation, upregulation of p21, and induction of apoptosis in non-small cell lung cancer (NSCLC) and drug-resistant cell lines. SNOH-3 also downregulates anti-apoptotic proteins XIAP and Survivin, inhibits tumor cell migration, invasion, and angiogenesis (partly by increasing KLF4 and reducing VEGF expression in endothelial cells). SNOH-3 demonstrates synergistic anti-tumor activity with paclitaxel, overcoming paclitaxel resistance in NSCLC models in vitro and in vivo. SNOH-3 exhibits greater or similar HDAC inhibitory capacity and pro-apoptotic activity compared to SAHA (vorinostat)[1].
