Overview

SNS-101 is a conditionally active, human monoclonal antibody designed to target the immune checkpoint protein V-domain Ig suppressor of T cell activation (VISTA). Developed by Sensei Biotherapeutics, SNS-101 is engineered using the company's TMAb (Tumor Microenvironment Activated) platform to selectively bind VISTA in the acidic environment (low pH) characteristic of the tumor microenvironment, while remaining inactive at the neutral pH of peripheral blood. This pH-selective binding is intended to mitigate the cytokine release syndrome (CRS) and rapid systemic clearance (target-mediated drug disposition) often associated with traditional VISTA-targeting agents. By blocking the VISTA-mediated suppression of T cells within the tumor, SNS-101 aims to restore anti-tumor immune responses. It is currently being evaluated in Phase 1/2 clinical trials for the treatment of advanced solid tumors, both as a monotherapy and in combination with PD-1 inhibitors like cemiplimab.
