SNS-103 is a conditionally active, monoclonal antibody targeting ecto-nucleoside triphosphate diphosphohydrolase 1 (ENTPDase1), also known as CD39. Developed by Sensei Biotherapeutics, it is designed to selectively bind and inhibit CD39 only under the acidic conditions found in the tumor microenvironment. CD39 is a rate-limiting enzyme responsible for the breakdown of extracellular ATP, leading to increased adenosine production and an immunosuppressive tumor microenvironment. By inhibiting CD39 activity specifically within tumors, SNS-103 aims to maintain extracellular ATP levels and reduce adenosine generation, thereby enhancing anti-tumor immunity while minimizing off-tumor effects. The drug remains in preclinical development for solid tumors[1][2][3][4][7][8].
