SNS-201

SNS-201 is a bispecific antibody developed by Sensei Biotherapeutics for oncology indications, specifically solid tumors and prostate cancer. It is designed as a conditionally active CD28xVISTA bispecific antibody, meaning it targets both the CD28 antigen (a T-cell co-stimulatory receptor) and VISTA (V-domain Ig suppressor of T cell activation), an immune checkpoint protein implicated in tumor immune evasion. The molecule consists of a CD28 agonist arm and a pH-sensitive anti-VISTA arm, enabling selective activity within the acidic tumor microenvironment. This dual targeting aims to stimulate T lymphocytes while simultaneously inhibiting VISTA-mediated immunosuppression, thereby enhancing anti-tumor immunity. Preclinical data have shown that SNS-201 can significantly inhibit tumor growth in relevant models[1][9]. As of early 2024, development was paused at the preclinical stage to prioritize other pipeline assets[1].
