SNS-314 is a potent and selective small molecule inhibitor of Aurora kinases A, B, and C. Aurora kinases are serine/threonine-protein kinases that regulate key processes in mitosis including centrosome maturation, mitotic spindle formation, chromosome alignment, and cytokinesis. Overexpression of these kinases is frequently observed in various human cancers such as colon, breast, ovarian, gastric, pancreatic tumors among others. By inhibiting all three Aurora kinases (AURKA IC50 ~9 nM; AURKB IC50 ~31 nM; AURKC IC50 ~3 nM), SNS-314 disrupts mitotic spindle assembly checkpoint leading to failure of cytokinesis and ultimately cell death through mechanisms like endoreduplication. This selective inhibition targets highly proliferative tumor cells while sparing most normal non-dividing cells. The drug was developed by Sunesis Pharmaceuticals primarily for treatment of advanced solid tumors and has been evaluated in Phase 1 clinical trials as a single agent with demonstrated pharmacodynamic activity and tolerability.
