Overview

SNS-510 is a first-in-class, orally bioavailable small molecule inhibitor of 3-phosphoinositide-dependent protein kinase 1 (PDK1, also known as PDPK1). PDK1 is a master kinase that regulates key signaling pathways involved in cell growth and survival, including the PI3K/AKT pathway. By binding to the inactive conformation of PDK1 and perturbing its PIF-pocket, SNS-510 inhibits both PI3K-dependent and PIP3-independent pathways critical for tumorigenesis. Preclinical studies have shown potent anti-proliferative activity in both solid tumors and hematologic malignancies, with particular sensitivity observed in cancer cell lines harboring CDKN2A mutations. The drug has demonstrated strong synergy with standard cancer therapies in preclinical models. Originally developed by Biogen and Millennium Pharmaceuticals (Takeda Oncology), it is being advanced by Sunesis Pharmaceuticals for oncology indications[1][4][5][7][8].
