**SNS01-T** is an investigational, intravenously administered polyethylenimine nanoparticle developed for relapsed or refractory B-cell malignancies. It co-delivers a B-cell-promoter-driven plasmid encoding the non-hypusinable, pro-apoptotic eIF5A K50R mutant and an siRNA that suppresses endogenous eukaryotic translation initiation factor 5A expression. The intended dual mechanism is depletion of pro-survival hypusinated eIF5A while expression of eIF5A K50R promotes apoptosis in malignant B cells. It entered a Phase 1/2 dose-escalation study in multiple myeloma, mantle cell lymphoma, diffuse large B-cell lymphoma, other B-cell lymphoma, and plasma cell leukemia; the sponsor subsequently placed the clinical program on hold.
