SNT-4288 is a lead oncology candidate developed by Sonata Therapeutics as part of its Network Medicines™ platform. It is a small molecule designed to induce ferroptosis, an iron-dependent form of regulated cell death, for the treatment of sarcomas. SNT-4288 acts as an inducer of ferroptosis, primarily by inhibiting glutathione peroxidase 4 (GPX4), an enzyme that neutralizes lipid peroxides and prevents oxidative cell death. By targeting the metabolic vulnerabilities of tumor cells, SNT-4288 aims to overcome resistance to conventional therapies. Preclinical data presented at the 2023 Society for Immunotherapy of Cancer (SITC) meeting demonstrated that SNT-4288 (also referred to as SNT-101) significantly reduces cell viability and induces lipid peroxidation in sarcoma models, showing potent anti-tumor activity and superior efficacy compared to the multi-kinase inhibitor pazopanib.
