Overview

SNTX-EA is a synthetic small molecule derivative of the naturally occurring compound SNTX-1675, developed by Sensorium Therapeutics using its Sens-AI platform. It is designed as a dual-action therapeutic to treat epilepsy and comorbid anxiety, addressing a significant unmet need in patients with drug-resistant epilepsy (DRE). SNTX-EA was optimized through a medicinal chemistry campaign to improve central nervous system (CNS) drug-like properties and pharmacokinetic (PK) exposure compared to its parent scaffold. In preclinical studies, the compound demonstrated anti-seizure efficacy across multiple rodent models, including the 6 Hz psychomotor (22, 33, and 44 mA), maximal electroshock seizure (MES), and corneal kindling assays. Additionally, it exhibited anxiolytic-like effects in marble-burying models without causing motor impairment on the rotarod. The molecule's mechanism involves the modulation of ion channels and CNS receptors, supporting its advancement toward clinical development for epilepsy with comorbid anxiety.
