SNV4818 is an orally administered, small molecule drug developed by Synnovation Therapeutics for the treatment of advanced solid tumors harboring activating PIK3CA mutations. It is a pan-mutant-selective inhibitor of phosphoinositide 3-kinase alpha (PI3Kα), specifically targeting common oncogenic mutations in both the kinase (H1047X) and helical (E545/542X) domains of PI3Kα while sparing wild-type PI3Kα. This selectivity aims to improve anti-tumor efficacy and reduce toxicities such as stomatitis, hyperglycemia, and rash that are associated with first-generation PI3Kα inhibitors. The drug is currently being evaluated in Phase I clinical trials as monotherapy and in combination with fulvestrant for patients with hormone receptor-positive/human epidermal growth factor receptor 2-negative advanced breast cancer and other solid tumors[2][5][6].
