SNX281 is a systemically active, intravenously administered small molecule agonist of the Stimulator of Interferon Genes (STING) protein. It was developed to promote anti-tumor immunity by activating the cGAS-STING pathway, leading to increased production of type I interferons and pro-inflammatory cytokines. This activation enhances both innate and adaptive immune responses against tumor cells. SNX281 is potent, specific for all prevalent human isoforms of STING, and induces rapid downstream signaling resulting in maturation and activation of antigen-presenting cells as well as robust T cell responses. Preclinical studies demonstrated complete regression of tumors in mouse models with a single intravenous dose; however, clinical development for solid tumors and lymphoma was terminated at phase 1 due to sponsor decision. The drug was originally discovered by Silicon Therapeutics using computational design platforms and is now wholly owned by Stingthera.
