SO1861 is a plant-derived glycosylated triterpenoid saponin isolated from the roots of *Saponaria officinalis* (soapwort). It functions as a potent endosomal escape enhancer, a technology designed to overcome the bottleneck of endolysosomal entrapment for macromolecular therapeutics. By transiently and specifically permeabilizing endosomal membranes, SO1861 facilitates the translocation of various "cargo" molecules—such as targeted toxins, antibody-drug conjugates (ADCs), and nucleic acids—from the endosome into the cytosol, where they can reach their intracellular targets. This synergistic approach can enhance the potency of targeted therapies by several thousand-fold while potentially reducing off-target toxicity. It is being developed as a key component of a delivery platform for oncology and other indications, often evaluated in combination with targeted biologics.
