SOA101 is a nanobody-based trispecific T cell engager (Nb-TriTE) designed to target and bind simultaneously to T cells, macrophages, and cancer cells. It redirects T cells toward tumor cells expressing the immune checkpoint proteins PD-L1 and/or HLA-G. The drug consists of nanobodies fused with a human IgG1 Fc domain, which prolongs its half-life. SOA101 enhances cytotoxicity mediated by human peripheral blood mononuclear cells (PBMCs), triggers T cell proliferation and activation, facilitates antibody-dependent cell cytotoxicity, and is intended to overcome the immunosuppressive tumor microenvironment by converting "cold" tumors into "hot" ones. Preclinical studies have shown broad-spectrum anti-tumor effects in vitro and superior anti-cancer potency compared to monoclonal antibodies or bispecific T cell engagers in non-small cell lung cancer models[1][3][4].
