Overview

TZT-1027, also known as soblidotin, is a synthetic derivative of dolastatin 10 and functions as an antimicrotubule agent. It exerts its antitumor effects by depolymerizing microtubules, leading to cell cycle arrest at the G2/M phase and inducing apoptosis in tumor cells. In addition to direct cytotoxicity against cancer cells, TZT-1027 exhibits unique anti-tumoral vascular activity by rapidly disrupting tumor vasculature and blocking blood flow within tumors. This dual mechanism results in both direct killing of tumor cells and indirect cytotoxicity through vascular blockade. TZT-1027 has demonstrated potent antitumor efficacy in various preclinical models, including activity against drug-resistant tumors, and has entered phase I clinical trials for solid tumors.
