Overview

Soblidotin + gemcitabine is an experimental combination chemotherapy regimen consisting of the dolastatin 10 derivative soblidotin and the nucleoside analog gemcitabine. Soblidotin (also known as TZT-1027) is a small molecule antimicrotubule agent that binds to beta-tubulin, inhibiting polymerization and inducing G2/M phase cell cycle arrest and apoptosis. Gemcitabine is a pyrimidine antimetabolite that inhibits DNA synthesis by acting as a nucleoside analog and inhibiting ribonucleotide reductase. The combination was investigated by Daiichi Sankyo in a Phase I clinical trial (NCT00072228) for patients with locally advanced or metastatic solid tumors. However, the trial was withdrawn before completion, and development of this specific combination has been discontinued.
