Overview

This is a combination of two small molecule drugs, sodium benzoate and N-acetylcysteine (NAC). Sodium benzoate acts primarily as a glycine scavenger by conjugating with glycine in the liver to form hippurate, which is then excreted in urine. This mechanism underlies its use in disorders such as non-ketotic hyperglycinemia and urea cycle disorders[5]. N-acetylcysteine is a precursor to glutathione, serving as an antioxidant and mucolytic agent. It also modulates glutamatergic neurotransmission via effects on cystine-glutamate exchange and has anti-inflammatory properties[6][7][8]. The combination has been studied for potential additive or synergistic effects on oxidative stress pathways, inflammation, NMDA receptor modulation, and neuroprotection—particularly in neuropsychiatric conditions like schizophrenia[1]. Both agents are well-tolerated individually; their combined use remains investigational.
