Drug intelligence / Profile preview
Sodium fluorocitrate (SFC) is a small-molecule research compound and the sodium salt of fluorocitrate, which is the active toxic metabolite of the pesticide sodium fluoroacetate (compound 1080). SFC acts as a potent, competitive inhibitor of the tricarboxylic acid (TCA) cycle enzymes mitochondrial aconitase (ACO2) and cytosolic aconitase (ACO1), halting cellular respiration and energy production. In preclinical research, low doses of SFC have been shown to protect pancreatic beta-cells and hepatocytes against palmitate-induced lipotoxicity by inhibiting cellular fatty acid uptake, an effect that appears independent of aconitase inhibition. In animal models of type 2 diabetes and high-fat-diet-induced non-alcoholic fatty liver disease (NAFLD), SFC treatment lowered blood glucose, reduced hepatic fat accumulation, decreased inflammatory signaling, and improved insulin resistance.
