Leucosodium is an injectable formulation of sodium folinate (also known as leucovorin sodium or folinate sodium), developed and manufactured by MyungMoon Bio. As a reduced folate derivative, it does not require enzymatic reduction by dihydrofolate reductase (DHFR), allowing it to maintain intracellular reduced folate pools even in the presence of DHFR inhibitors like methotrexate. Leucosodium is approved as a rescue therapy to prevent or reduce the severe toxicities associated with high-dose methotrexate administration in osteosarcoma and other neoplastic diseases. It is also widely used in combination with 5-fluorouracil (5-FU) to treat advanced colorectal cancer, where its active metabolite (5,10-methylenetetrahydrofolate) stabilizes the binding of 5-FU to thymidylate synthase, leading to prolonged enzyme inhibition and enhanced antineoplastic efficacy.
