SOF-XX is a preclinical-stage ultra-short oligonucleotide (USO) developed by Pharmorage, specifically engineered to target and antagonize Toll-like receptor 7 (TLR7). USOs represent a novel therapeutic modality characterized by their extremely short sequence length (typically 2 to 5 nucleotides), which distinguishes them from traditional antisense oligonucleotides or aptamers. By inhibiting TLR7, SOF-XX prevents the activation of downstream signaling pathways that lead to the overproduction of type I interferons and other pro-inflammatory cytokines. This mechanism is particularly relevant for the treatment of systemic lupus erythematosus (SLE), psoriasis, and various inflammatory skin diseases, where TLR7 signaling is a key driver of chronic inflammation and autoimmunity. The USO platform is designed to offer improved stability and delivery profiles compared to longer oligonucleotide sequences.
