Overview

Sofalcone is a synthetic flavonoid and a derivative of sophoradin, originally isolated from the root of the medicinal plant *Sophora subprostrata*. It is classified as a chalcone and functions primarily as a mucosal protective agent for the gastrointestinal tract. Sofalcone is approved in Japan and South Korea for the treatment of gastritis, gastric ulcers, peptic ulcers, and other gastric mucosal lesions. Its mechanism of action includes enhancing mucosal defense by inhibiting prostaglandin degradation enzymes, directly inhibiting growth and adhesion of *Helicobacter pylori*, exhibiting anti-urease activity against this pathogen, reducing production of vacuolating toxin (VT), suppressing interleukin-8 (IL-8) secretion induced by *H. pylori*, and exerting anti-inflammatory effects through covalent targeting of high mobility group box 1 (HMGB1) protein in intestinal epithelial cells[1][2][3][5][6][7][8].
