Overview

**Sofetabart mipitecan (LY4170156)** is a novel **antibody-drug conjugate (ADC)** developed by **Eli Lilly and Company** that targets **folate receptor alpha (FRα)**, a protein overexpressed on ovarian cancer cells. It consists of a monoclonal antibody linked to an **exatecan payload**, a topoisomerase I inhibitor, enabling targeted delivery of the cytotoxic agent to FRα-expressing tumors, potentially improving efficacy and reducing off-target toxicity compared to systemic chemotherapy. It is currently being evaluated in the phase 3 **GOG-3133/FRAmework-01** trial for **platinum-resistant** and **platinum-sensitive recurrent ovarian cancer**, including peritoneal and fallopian tube cancers, with part A comparing it to chemotherapy or mirvetuximab soravtansine, and part B assessing it plus bevacizumab versus platinum-based chemotherapy plus bevacizumab.[1][11]
