Drug intelligence / Profile preview
Sofe-M + bevacizumab is an investigational combination regimen consisting of sofetabart mipitecan (Sofe-M; LY4170156), a folate receptor alpha (FRα)-targeting antibody-drug conjugate (ADC), and bevacizumab, an anti-vascular endothelial growth factor (VEGF) monoclonal antibody. Sofetabart mipitecan is composed of an Fc-silent, humanized IgG1 monoclonal antibody targeting FRα, conjugated to the topoisomerase I inhibitor payload exatecan via a proprietary cleavable polysarcosine linker (PSARlink) with a drug-to-antibody ratio (DAR) of 8:1. Bevacizumab is a recombinant humanized monoclonal antibody that binds to and neutralizes VEGF, inhibiting angiogenesis. This combination is being developed by Eli Lilly and Company for the treatment of ovarian cancer, specifically platinum-sensitive ovarian cancer (PSOC), where it has demonstrated promising clinical efficacy and tolerability in Phase 1 trials, and is currently being evaluated in the Phase 3 FRAmework-01 master protocol.
