Sofituzumab vedotin is an investigational antibody-drug conjugate (ADC) developed by Genentech/Roche for the treatment of cancers that overexpress MUC16, such as ovarian and pancreatic cancer. The drug consists of a humanized anti-MUC16 (CA125) monoclonal antibody linked to the cytotoxic agent monomethyl auristatin E (MMAE). Upon binding to MUC16 on tumor cells, the ADC is internalized and releases MMAE, which inhibits tubulin polymerization, leading to cell cycle arrest and apoptosis. Additional antitumor effects may include signal transduction inhibition, as well as immune-mediated mechanisms like antibody-dependent cellular cytotoxicity and complement-dependent cytotoxicity[1][3][4][6].
