Overview
Sofosbuvir is a direct-acting antiviral medication used in combination with other agents to treat chronic hepatitis C virus (HCV) infection in adults and children aged 3 years and older. It is a nucleotide analog inhibitor that targets the HCV NS5B RNA-dependent RNA polymerase, an enzyme essential for viral replication. Sofosbuvir acts as a prodrug, which is metabolized intracellularly to its active triphosphate form (GS-461203). This active metabolite incorporates into HCV RNA by the NS5B polymerase, resulting in chain termination and inhibition of viral replication. Sofosbuvir was developed by Gilead Sciences and represents a significant advance in hepatitis C therapy due to its high efficacy, oral administration, and improved safety profile compared to previous treatments[1][2][3][5][7].
