Overview

Sofosbuvir + daclatasvir + ribavirin is an oral combination regimen used for the treatment of chronic hepatitis C virus (HCV) infection, particularly in patients with genotype 3 and those with advanced liver disease or cirrhosis. Sofosbuvir is a nucleotide analog inhibitor of the HCV nonstructural protein 5B (NS5B) RNA-dependent RNA polymerase, which inhibits viral replication. Daclatasvir is a pangenotypic inhibitor of the HCV nonstructural protein 5A (NS5A), essential for viral replication and assembly. Ribavirin is a nucleoside analog with broad-spectrum antiviral activity that enhances virologic response when combined with direct-acting antivirals. This triple therapy has demonstrated high sustained virological response rates in clinical trials, especially among difficult-to-treat populations such as those with cirrhosis or prior treatment failure[2][3][6]. The regimen is generally well tolerated; common adverse effects include headache, fatigue, nausea, insomnia, and hemolytic anemia[2][3][6]. The combination should not be used as monotherapy due to risk of resistance development.
