Overview

A combination of three antiviral agents used in clinical trials for the treatment of chronic hepatitis C virus (HCV) infection, especially genotype 1. **Sofosbuvir** is a nucleotide analog inhibitor of the HCV NS5B RNA-dependent RNA polymerase. **Ribavirin** is a nucleoside analog with broad-spectrum antiviral activity, often used to enhance efficacy in combination regimens. **GS-9669** is a non-nucleoside inhibitor of the HCV NS5B polymerase (thumb site II inhibitor) with activity against genotype 1. This combination was studied as an **interferon-free, all-oral regimen** aimed at achieving high rates of sustained virologic response (SVR) in treatment-naive and treatment-experienced patients, including those with cirrhosis. GS-9669 specifically binds the thumb site II of NS5B, causing conformational changes that inhibit viral RNA synthesis[2][4]. The regimen was well-tolerated in trials, with adverse events mainly including headache, nausea, and fatigue[1][3].
