Solitomab is a bispecific monoclonal antibody of the BiTE (bispecific T-cell engager) class. It is designed to simultaneously bind to the epithelial cell adhesion molecule (EpCAM) on tumor cells and CD3 on T cells. By physically linking T cells to EpCAM-expressing tumor cells, solitomab redirects cytotoxic T lymphocytes to kill cancer cells through perforin- and granzyme-mediated apoptosis, independent of MHC I or co-stimulatory molecules. The drug was originally developed by Micromet and later by Amgen after acquisition. Its primary indication has been for various solid tumors expressing EpCAM, including gastrointestinal cancers, lung cancer, ovarian cancer, breast cancer, colorectal cancer, gastroesophageal junction adenocarcinoma, castration-resistant prostate cancer, small cell lung cancer, and uterine carcinosarcoma[2][5][7][9].
