Solnerstotug is a conditionally active, pH-selective monoclonal antibody that targets the immune checkpoint VISTA (V-domain Ig suppressor of T-cell activation). It is designed to restore T cell activity in patients with advanced solid tumors, particularly those who have become resistant to prior PD-(L)1 immunotherapy. By antagonizing VISTA, solnerstotug aims to overcome immune suppression within the tumor microenvironment and enhance anti-tumor immunity. The drug is being developed by Sensei Biotherapeutics and has shown promising preliminary efficacy and safety data in early-phase clinical trials, both as monotherapy and in combination with cemiplimab (a PD-1 inhibitor), especially for patients with "hot" tumors that are typically responsive to immunotherapy but have progressed on standard treatments[2][3][5][7].
