Drug intelligence / Profile preview
Soluble Nogo-B (sNogo-B) is a circulating, soluble N-terminal fragment (~150–200 amino acids) of the endoplasmic reticulum-associated protein Nogo-B (Reticulon 4B). It is being investigated as a potential therapeutic agent for diabetic nephropathy (diabetic glomerulopathy) and other diabetic vascular complications. sNogo-B acts as a ligand for the Nogo-B receptor (NgBR), promoting endothelial cell survival, migration, and vascular integrity. In preclinical models of diabetic kidney disease, systemic overexpression or administration of sNogo-B has been shown to protect the glomerular filtration barrier, reduce albuminuria and hyperfiltration, restore the endothelial glycocalyx, and normalize aberrant angiogenesis by modulating VEGF-A/VEGFR2, Akt, and GSK3β signaling pathways.
