Soluble siglec-9 is a recombinant, soluble form of the human sialic acid-binding immunoglobulin-like lectin 9 (Siglec-9), an immunoregulatory protein of the Ig superfamily. As a decoy receptor, soluble siglec-9 binds to sialic acid–containing ligands and modulates immune function by antagonizing endogenous, cell-bound siglec-9 signaling. Preclinical studies show that this protein can inhibit pro-inflammatory responses in models of inflammatory bowel disease and rheumatoid arthritis, primarily by suppressing the activation of nuclear factor kappa B (NF-κB) in immune and epithelial cells, reducing cytokines such as TNF-α and IL-8, and rebalancing regulatory and effector T cell subsets. Soluble siglec-9 acts as an immune checkpoint modulator and is being investigated as a potential therapeutic agent in autoimmune and inflammatory diseases.
