Soluble TGFβ type III receptor (also known as soluble betaglycan or sRIII) is a recombinant soluble protein consisting of the extracellular domain of the transforming growth factor-beta (TGF-β) type III receptor. It acts as a high-affinity decoy receptor (ligand trap) that selectively binds and sequesters TGF-β isoforms, particularly TGF-β1, TGF-β2, and TGF-β3, thereby preventing them from interacting with their signaling receptors (TGFBR1 and TGFBR2). Preclinical studies have demonstrated that soluble TGFβ type III receptor can suppress tumor growth, angiogenesis, and metastasis in various cancer models, including breast cancer and breast cancer-induced osteolytic bone metastasis, by antagonizing the tumor-promoting activities of autocrine and paracrine TGF-β.
