SoluFlex Link ADC

SoluFlex Link ADC refers to a class of antibody-drug conjugates (ADCs) utilizing the SoluFlex Link, a novel hydrophilic solubilizing linker platform designed to improve the therapeutic index of hydrophobic payloads. Developed to address common ADC challenges such as poor manufacturability, reduced stability, and suboptimal pharmacokinetics, the SoluFlex Link increases the overall hydrophilicity of the conjugate beyond that of the payload itself. This platform has been evaluated with various antibodies and payload classes, including topoisomerase I inhibitors (such as exatecan) and attenuated DNA alkylators. In preclinical studies, SoluFlex Link-based ADCs demonstrated improved thermal stability and significantly enhanced in vitro potency, shifting IC50 values from micromolar to low-nanomolar ranges while maintaining target specificity. Notably, exatecan-based ADCs using this linker have shown superior tumor growth inhibition and regression rates compared to trastuzumab deruxtecan (T-DXd) in tolerated dose ranges.
