Overview

SomatoKine is a recombinant fusion protein developed by Insmed (formerly Celtrix Pharmaceuticals). It combines insulin-like growth factor-1 (IGF-1) with its binding protein BP-3, creating a longer-acting form of IGF-1[1][4]. The drug was developed primarily for treating growth hormone insensitivity syndrome (GHIS), a condition where patients cannot respond properly to growth hormone. SomatoKine was also being investigated as a potential injectable insulin sensitizer for managing both type 1 and type 2 diabetes in patients with reduced insulin sensitivity[1]. By July 2002, the FDA had granted SomatoKine Orphan Drug status for GHIS treatment. Around that time, a phase II clinical trial in children with GHIS was initiated. In September 2002, Insmed planned to start another clinical trial for GHIS in early 2003 and continue phase II development for diabetes[1]. One of the key advantages of SomatoKine's design was to prolong the half-life of IGF-1 and to reduce acute adverse events associated with IGF-1 administration, particularly hypoglycemia[4]. ## Development Status As of the search results provided, SomatoKine was in clinical development. Reduce the risk of hypoglycemia compared to IGF-1 alone by creating a more stable delivery system[1][4] Unlike somatropin (recombinant human growth hormone), which acts earlier in the growth hormone pathway, SomatoKine provides the downstream effector (IGF-1) directly, making it useful for conditions where growth hormone receptors don't function properly.
