Overview

This is a combination of two drugs, somatostatin and octreotide. Both are peptide hormones that act as inhibitors of various endocrine, neuroendocrine, exocrine, and paracrine functions. Somatostatin is an endogenous hormone that inhibits the secretion of several other hormones such as growth hormone (GH), insulin, glucagon, gastrin, and others. Octreotide is a synthetic long-acting analogue of somatostatin with similar but more potent inhibitory effects on hormone secretion. The combination has been studied for synergistic or additive effects in conditions where inhibition of pancreatic or gastrointestinal secretions is desired. Mechanistically, both drugs bind to somatostatin receptors (primarily SSTR2 and SSTR5), leading to decreased release of target hormones and peptides from various tissues. This results in reduced gastrointestinal motility and secretions as well as inhibition of tumor-related hormonal syndromes. The combination has been investigated particularly in acute pancreatitis for its potential to reduce complications by suppressing pancreatic enzyme secretion more effectively than either agent alone[5]. Both agents are also used individually for the management of acromogaly, carcinoid syndrome symptoms (such as diarrhea/flushing), vasoactive intestinal peptide-secreting tumors (VIPomas), and neuroendocrine tumors[2][3][4][6][7].
