Somatotropin is a recombinant form of human growth hormone (hGH), a protein naturally produced by the pituitary gland. It is used to treat children and adults with growth hormone deficiency and various conditions that result in short stature or impaired growth, such as Turner syndrome, Prader-Willi syndrome, chronic kidney disease, Noonan syndrome, SHOX deficiency, idiopathic short stature, and children born small for gestational age who fail to catch up in growth. In adults, it is indicated for adult-onset or childhood-onset growth hormone deficiency due to pituitary or hypothalamic disease. Somatotropin is also approved for HIV-associated wasting/cachexia and short bowel syndrome in adults receiving specialized nutritional support. Mechanistically, somatotropin binds to the human growth hormone receptor (GHR) on target cells—primarily in the liver and cartilage—activating JAK2/STAT signaling pathways that stimulate gene transcription involved in linear bone growth and metabolic regulation. Its effects are mediated both directly via GHR activation and indirectly through increased production of insulin-like growth factor 1 (IGF‑1), which further promotes tissue anabolism[5][7][8].
