Somatrogon is a long-acting recombinant human growth hormone used as hormone replacement therapy to treat pediatric patients aged 3 years and older with growth failure due to inadequate secretion of endogenous growth hormone. It is a chimeric protein produced using recombinant DNA technology in Chinese Hamster Ovary (CHO) cells, consisting of the amino acid sequence of human growth hormone fused to three copies of the C-terminal peptide from the beta chain of human chorionic gonadotropin. This modification extends its half-life, allowing for once-weekly dosing. Somatrogon binds to the growth hormone receptor, activating JAK2 and STAT5b signaling pathways, which stimulate metabolic changes and promote linear growth by increasing serum IGF-1 levels[1][3][6][8]. The drug was jointly developed by Pfizer and OPKO Health.
