Overview

SON-1010 is a recombinant immunotherapeutic drug candidate developed by Sonnet BioTherapeutics. It consists of unmodified single-chain human interleukin-12 (IL-12) genetically fused to an albumin-binding domain derived from the single-chain antibody fragment A10m3, using Sonnet’s Fully Human Albumin Binding (FHAB) technology. This design enables targeted delivery of IL-12 to tumor and lymphatic tissues by exploiting albumin’s natural transport pathways and its affinity for proteins overexpressed in the tumor microenvironment (TME), such as SPARC and GP60. The mechanism aims to enhance immune activation within tumors—turning ‘cold’ tumors ‘hot’—by stimulating interferon gamma (IFNγ) production, which activates both innate and adaptive immune responses and increases PD-L1 expression on tumor cells. SON-1010 is being investigated primarily for advanced solid tumors, including non-small cell lung cancer, melanoma, head and neck cancer, sarcoma, gynecological cancers, and platinum-resistant ovarian cancer[1][3][4][5][6][8].
