SON-1210 is an investigational bifunctional immunotherapeutic fusion protein that combines unmodified single-chain human interleukin 12 (IL-12) and human interleukin 15 (IL-15), separated by the albumin-binding domain of a single-chain antibody fragment known as FHAB. This design allows targeted delivery of both cytokines to tumor and lymphatic tissues, leveraging the acidic pH and protein environment of the tumor microenvironment (TME). The FHAB technology enhances pharmacokinetics, increases tumor retention, and may improve safety and efficacy compared to non-modified cytokines. Mechanistically, SON-1210 aims to stimulate interferon gamma (IFNγ) production in tumors, activating both innate and adaptive immune responses—effectively turning immunologically "cold" tumors "hot." It is being developed primarily for solid tumors including pancreatic cancer, colorectal cancer, non-small cell lung cancer, melanoma, head and neck cancers, sarcoma, and certain gynecological cancers. The drug is currently in preclinical development with plans for Phase 1/2a clinical trials in metastatic pancreatic cancer[1][3][5][7][10].
